Genomes of Anguillid Herpesvirus 1 Strains Reveal Evolutionary Disparities and Low Genetic Diversity in the Genus Cyprinivirus
Round 1
Reviewer 1 Report
This manuscript entitled “Genomes of Anguillid herpesvirus 1 strains reveals evolutionary disparities and low genetic diversity in the genus Cyprinivirus” reported interesting data about genetic diversity of AngHV-1. A strong point of the study was the isolation and the whole genome sequencing of 7 AngHV-1 strains collected from various geographical origins to investigate viral genetic diversity and the biological features of AngHV-1. Indeed, very few genomes of AngHV-1 were whole sequencing and there is no available data about biological properties of them.
Introduction section clearly presented the context and the aim of the study. Material and method section are particularly well detailed. However, viral plaque area assay could be improved by adding some references related to the immunodetection method. Results were also clearly presented.

Reply: Thank you, the methods section has been updated appropriately (Line 136-148 changes tracked and highlighted in green).
Reviewer 2 Report
This study analyses genomes of auguillid herpesvirus 1 of multiple geographical origins, discloses siginificantly less genetic diversity in core gene sequences within cyprinivirus clades compared to species in the family Herpesviridae, and discuss the base of the difference. The manuscript is well constructed and written, requiring no revision except for a few points where the reviewer requests checking by the authours. These points are mentioned below.
- L96-L97 [Table 1]: References shown in Table 1 should be listed in the References section, and replaced by the reference numbers or added these information.
- L96-L97 [Table 1]: What are ‘the first seven AngHV-1 strains’ in the footnote?
- L151: ‘BLASTX’
- L199, L249, L293, L309, L311, L313, L438, L471, L473, L475, P710, P712: The letter ‘p’ for ‘p-value’ might be italicized for easy understating.
- L257: Is it correct to mention ‘three geographic locations’?
- L443: Is it correct to place ‘may be’ before ‘have been deriven’? ‘maybe’ or ‘may have been deriven’ (?)
- L485: ‘as being driven …’ (?)
