Special Issue Information

Dear Colleagues,

Brain metastases are the most prevalent form of brain malignancy associated with poor prognosis and reduced quality of life. There is a need to investigate different mechanisms and novel treatment approaches to target brain metastasis. The process of brain metastasis involves complex crosstalk between diverse cell types and processes. The mechanisms including the interaction with neurotransmitters and growth factors in the brain microenvironment, vascular co-option and the modification of the blood–brain barrier contribute to disease progression and treatment refractory. Advancements in single-cell omics and multiplex spatial imaging promise a better understanding of the metastatic brain ecosystem, offering valuable insights for potential treatment strategies. Challenges in drug delivery across the blood–tumor barrier and the brain tumor microenvironment have led to poor control of brain metastasis. Overcoming these challenges and improving treatment efficacy with minimal systemic toxicity using advanced approaches is pivotal.

This Special Issue focuses on the molecular intricacies in brain metastasis and the novel vulnerabilities in the brain microenvironment for targeted therapeutic interventions. It explores the dynamic interplay of brain resident cells, immune cells, neurotransmitters, and metabolic adaptations to identify therapeutic targets. This Special Issue also aims to compile studies exploring advanced therapeutic approaches for treating brain metastases. We welcome both original research articles and comprehensive reviews to contribute to this collective effort.
