In this review paper, the authors have comprehensively reviewed the literature on how colorectal cancer cells invade and cause atrophy of the enteric nervous system.  The authors have discussed the role of growth factors and galanin in the invasion of cancer cells locally and in the surrounding tissues.  They have discussed the role of galanin in elucidation of signal transduction pathways and how it inhibits extrinsic pathway of apoptosis and promotes cancer cell proliferation.  They also discussed the role of neurotrophic factor in cancer cell proliferation. 

Minor comments:

  1. A model figure diagrammatically representing the various signaling pathways activated by growth factors and galanin contributing to the invasion and entropy of the tssues is required in this review.
  2. Throughout the text I noticed spelling errors. Please correct.  An example is on page 2, last paragraph, please change the word “Neoplasmic” to “Neoplastic”.

Manuscript of a review entitled "Colorectal cancer invasion and atrophy of the enteric nervous system – potential feedback and impact on cancer progression" by Godlewski and Kmiec. The readability of this manuscript could be improved a lot with illustrations and tables. For example an illustration of what is known about different neuropeptides in colorectal cancer progression. As it is current state, it is mainly concentrating on one neuropeptide, galanin, but other should be taken along to present a good comparison for readers to evaluate. Also, a comprehensive table of neuropeptides and neurotropic factors in colorectal cancer versus acute inflammation of the colon, would be a useful addition. There are grammar mistakes in the text as well as some sentences that are hard to understand, which should be corrected.

Thank you valuable and helpful suggestions. Indeed, we prepared a Table which presents neuropeptides’ function in normal large intestine and their possible effects/function in CRC cell lines, tumuor tissue and in experimentally-induced tumours.

Figure 1 entitled “Function and localization of the ENS neurotransmitters in the normal human large intestine and colorectal cancer cell lines and tumour tissue” has been added in response to your comment.

We apologize for low English-editing level of the original version, we hope that the corrected MS is much better readable.
