The Use of Diffusion Kurtosis Imaging for the Differential Diagnosis of Alzheimer’s Disease Spectrum
Round 1

Comments and Suggestions for AuthorsThe article is good written and illustrated, however, I have two remarks:
1. Figure 3 is cut on the left side, so I couldn t see the left part of it.
2. My main question/comment is about ROI choice. As I understood from the methods, for volume quantification the authors used ROI from CAT12 program, and for DKI - ROI from AAL alas in FSL. Are these ROI complimentary and match? Are the authors sure they have measured GM, and no surrounded white matter for AAL ROI - otherwise presence of WM in ROI can influence on results. And it would be nice to illustrate the hippocampus ROI from CAT in Figure 1 (by the way, what it the difference and purpose for putting results of 2 subjects (?) illustration in Figure 1?)

Comments and Suggestions for AuthorsThis topic is interesting and current. Look at some points to improve it:
- The title does not reflect the whole paper. Better it can be: "The use of Diffusion Kurtosis Imaging for the Differential Diagnosis of Alzheimer's Disease Spectrum"
- "In the preclinical SCD stage.. mediators such as proinflammatory cytokines and chemokines [16]" Is there any relation to parkinson's disease? Discuss about it: --- doi: 10.1007/s00281-022-00944-6 --- doi: 10.3390/ijerph19148799 --- doi: 10.1007/s12035-020-01926-1
- "Our result showed that hippocampal atrophy became predominant at the AD stage, where volume loss is much more evident than the microstructural changes [19,47]." What do authors think about this?
- Figure 6 is too big, try to merge it.
- improve conclusion. What does this paper add new?
