Curated Dataset for Red Blood Cell Tracking from Video Sequences of Flow in Microfluidic Devices
Round 1
Reviewer 1 Report
1. Line 171-172 - confusing usage of multiply sign (x) with resolution unit (px)
2. Some units are in italic, some are not - please be consistent
3. Fig. 3 - please be specific on the '1st video' referred. It is suggested to name the video set rather than calling as video 1 and video 2
4. Fig. 4 - spelling (slit)
5. Table 1 - what are the units?
6. References - incomplete citation (5, 6, 16, 20-22, 26)
The writing of the paper in overall is good. Language is good. Only the conclusion part is missing.

The manuscript reports dataset of images, annotations, and velocity fields for cell detection and cell tracking algorithms. The flow of red blood cells (RBC) in microfluidic channels were recorded and a conventional image processing algorithm to detect the centers of the cells and tracking were applied. The dataset contains two videos, one contains a recording of flow of a cell suspension in an U-shaped channel and another recording of the flow of cell suspension in a channel that is constricted by four horizontal obstacles creating three narrow slits with different widths. Both videos are annotated, the first containing tracks with 176.9 frames on average and the second containing tracks with 18.8 frames on average. Both videos have a completely different behavior of 178 cells. The dataset contains annotations of bounding boxes around the cells and information about the cell tracks. I think the manuscript presents interesting results in a well-structured text. The sentence “One of the biggest challenges in microbiology is collecting data” in the introduction does not make sense since the data presented is about mammalian cells.

This work titled with “Curated dataset for red blood cell tracking from Video sequences of flow in microfluidic devices”, is an interesting work with some novelty. This reviewer thanks it could be accepted for this journal with minor revision:
1. How to recognize, dismiss or handle with the fake cells (cell debris, contaminates or the images with low pixel-resolutions) ?
2. Any reprints from CoRR or bioRxiv could not be accepted as references. The authors are encouraged to check all the references and replace them with the publications.
3. As well, the authors are encouraged to check all the styles of references according to the requirement of this journal. “pp” from Ref 1-2,6,9,19 and 37-40 is not correct.
4. Statistical analysis (e.g. application codes, software version and methodology) should be present.
5. There are too many short paragraphs with only 1-2 sentences. Kindly restructure the full context and present more in Discussion and a conclusion section in the end of this work.
