Targeting Cancer Metabolism for the Development of Diagnostic, Therapeutic and Theranostic Agents

Dear Colleagues,
Cancer cells reprogram metabolic and nutrient acquisition paths to satisfy biosynthetic, redox, and bioenergetic demands to support survival, accelerated cell proliferation, and adaptation to the tumor microenvironment. Almost one hundred years have passed since Otto Warburg revealed that an altered metabolism in cancer cells influences their growth. Although cancer metabolism is considered to be one of the oldest research areas in cancer biology and a hallmark of cancer, it has not been fully exploited or explored for the development of novel and effective cancer therapeutic, diagnostic, or theranostic strategies. In order for this to be achieved, orchestrated action, on a system-wide scale, is required for cancer metabolome and proteome cartography through state-of-the-art analytic techniques. Furthermore, we need novel chemical biology tools in order to interrogate the metabolic reprogramming in tumor cells and the mechanistic details that connect the altered cancer cell metabolism to metastasis and tumorigenesis.
How metabolism is reprogrammed in cancer cells, how this can be exploited for the development of therapeutics, diagnostics, and theranostics, and how cancer-altered biochemical paths can be probed will be among the topics that will be covered in this Special issue. Focus will also be placed on multimodal imaging agents (for MRI, in vivo near-infrared I and II optical imaging, SPECT/PET, and photoacoustics) that target and respond to the tumor microenvironment of cancer cells or report on the reprogrammed metabolic pathway of cancer cells.
