1. In the introduction authors raised the problem of proteoglycans for RNA extraction. However I found no information further about neither any obstacles they impose on the RNA extraction, nor any proof of their successful elimination.
2. In discussion line 222-223 authors begin to speculate on how column extraction influence the purity of the extracted RNA, but the next sentence appears to be about effective lysis and protection from degradation. These two sentences seem little connected to each other.

This study entitled “Isolation of the “RNA-seq-quality” RNA from human intervertebral discs after mortar and pestle homogenization” seems to be well executed and written paper. Furthermore, I believe that this paper will be of great interest to the readers. Finally, I have only few minor suggestions to further improve the quality of this very interesting and scientifically significant paper.

Please begin this section with an information what type of study you have performed, in which time period and where. Following that sentence please state that this work was approved by the Local Ethical Committee of the Novosibirsk Research Institute of Traumatology and Orthopedics (please include the date) and that all patients provided written informed consent, as you stated below the text.

At the end of section Materials and Methods please add the subsection Statistical analysis to describe the statistical methods that you have performed in your research, and please state in which statistical program they have been executed.
