Look at these points:

- Lines 40-44: ""The primary objective of this review is to provide... a therapeutic target for various neurological disorders" Improve this part. What is the purpose of this review. What are the primary objectives?

- Is this a narrative review or a systematica review? State this, and accordingly, revise the paper.

- Lines 83-141. "3. Overview of Neuroglial Cells" I'm not sure this paragraph is necessary. What is the purpose of this paragraph?

- 172 references are too much, are all these references necessary?

- Lines 484-486: "Emerging pharmacological interventions targeting the microbiome-neuroglia axis offer promising therapeutic strategies for neurological disorders. " Discuss this more

Lines 40-44: ""The primary objective of this review is to provide... a therapeutic target for various neurological disorders" Improve this part. What is the purpose of this review? What are the primary objectives?

Response:

We now state in the abstract that his scoping review aims to highlight the mechanisms of the microbiome-neuroglia axis in maintaining brain health and its potential as a therapeutic target.

Comment:

Is this a narrative review or a systematic review? State this, and accordingly, revise the paper.

Response:

We specified in the abstract that this is a scoping review.

Comment:

Lines 83-141. "3. Overview of Neuroglial Cells" I'm not sure this paragraph is necessary. What is the purpose of this paragraph?

Response:

It aims to establish a foundational understanding of glial cells, highlighting their importance in maintaining CNS homeostasis, supporting neuronal function, and responding to pathological conditions. This context is essential for understanding subsequent discussions about the interactions between neuroglia and the gut microbiome, as well as their implications for brain health and neurological disorders.

Comment:

172 references are too much, are all these references necessary?

Response:

The high number of references is intended to provide a comprehensive and well-supported review of the current state of research on the microbiome-neuroglia axis and its implications for brain health and neurological disorders. Each reference contributes to the robustness and credibility of the review, ensuring that it accurately reflects the breadth and depth of existing scientific knowledge on this complex and rapidly evolving topic.

Response:

As the reviewer suggested, we provided additional details. Changes are highlighted in yellow.

Comments and Suggestions for Authors

Alberto K. De la Herrán-Arita submitted a review entitled The Gut Microbiome-Neuroglia Axis: Implications for Brain Health, Inflammation, and Disease. The authors comprehensively analyze the microbiome-neuroglia axis and its implications for brain health and neurological disorders. They aim to highlight the critical role of this axis in maintaining brain health and its potential as a therapeutic target for various neurological disorders. The paper is well prepared and may be interesting for the readers. The figures are of good quality. I want to ask if the figures are original. If not, the authors should include the note „adopted from…” Sometimes the agree is needed.

Author Response

We appreciate the reviewer's insightful suggestions and have made our best effort to address their concerns comprehensively.

As the reviewer suggested, we added “This figure was made in BioRender” in the figure legends. Changes are highlighted in yellow.

Comments and Suggestions for Authors

The manuscript, “The Gut Microbiome-Neuroglia Axis: Implications for Brain Health, Inflammation and Disease” by Dr. Alberto K. De la Herrán-Arita, aimed to review the

microbiome-neuroglia axis, exploring its implications for normal and pathological brain functioning and some neural disorders. Author observed the gut microbiome's influence on neural system through microbial metabolites and some other phenomena, including but not limited the modulation of the immune system, and neuroinflammatory responses.

This topic is undoubtedly extremely attractive and many scientific groups are working in this direction. The manuscript provides a serious analysis of the recent advancement in the field.

I have no fundamental objections, but I have some questions.

The author writes about hypothalamic-pituitary-adrenal axis and its regulation. Some materials are provided on how various pathways regulate gut motility, secretion, and microbial on the part of the CNS using neurotransmitters such as acetylcholine. These pathways are quite complex and diverse, and some data on the involvement of Nicotinic and Muscarinic acetylcholine receptors were obtained relatively recently. I would advise the author to add new data on this topic, for example using Tizabi et al, 2024 (doi: 10.3390/brainsci14060558 ), and perhaps some other modern publications.

Minor criticism: The manuscript lacks Abstract and Keywords. If it's allowed by the journal's rules, I don't mind, but it's a little unusual.

The manuscript is organized well, and written clearly, I was impressed very much. I will be happy to recommend the manuscript for the publication after minor correction, suggested before.

We have incorporated the data on the involvement of Nicotinic and Muscarinic acetylcholine receptors. Additionally, we have now included an abstract and keywords as per your recommendation. We appreciate your thorough review and constructive suggestions to improve the manuscript. Changes are highlighted in yellow.
