Neural stem cells (NSCs) hold immense potential for advancing our understanding and treatment of neurological diseases. Their ability to self-renew, differentiate into various neural cell types, and recapitulate disease-specific phenotypes makes invaluable tools for use in disease modeling. This multidisciplinary frontier involves utilizing NSCs obtained from diverse sources, including embryonic stem cells, induced pluripotent stem cells, and adult neural tissues, to replicate the complexity of neurological disorders in vitro. By manipulating NSCs, researchers can generate three-dimensional organoids or two-dimensional culture systems that mimic the characteristics of neurodegenerative diseases, neurodevelopmental disorders, and other conditions affecting the nervous system. These NSC-based models enable the investigation of disease mechanisms, drug screening, and the development of personalized therapies. Furthermore, NSCs can be genetically modified to express disease-specific mutations, providing a powerful platform for understanding the molecular and cellular basis of various neurological conditions. This Special Issue illuminates recent advancements in NSCs utilization to model neurological diseases, highlighting their potential to unravel disease pathogenesis and facilitate the development of new therapeutic strategies.
