Dear Colleagues,
Despite the major and constant efforts of the past 20 years, patients suffering from glioblastoma (GBM), the most frequent and deadly primary brain tumor, still have a very poor prognosis. Despite standard treatments combining surgical resection, when possible, radiochemotherapy and, recently, tumor-treating fields, these GBMs almost invariably recur. This rapid relapse is notably due to the diffuse infiltrative growth of GBM cells in the surrounding brain tissue, the pronounced intratumoral heterogeneity, the presence of GBM stem cells, a high tumor cell plasticity, a strong propensity to immune escape, and a high intrinsic resistance of GBM to cell death. In light of numerous recent fundamental and preclinical studies, significant progress has been made in understanding the complex molecular processes underlying gliomagenesis and treatment resistance in GBM, identifying a number of potential new targets and therapeutic strategies for the treatment of these highly aggressive brain tumors. The new engaging therapeutic options aim, in combination with conventional therapy, to exploit the specific flaws of GBM via high-precision treatments and possibly through personalized approaches. We welcome original research papers and review articles focusing on recent advances in this challenging area of research.
