Soybean Protein Extraction by Alcalase and Flavourzyme, Combining Thermal Pretreatment for Enteral Feeding Product
Round 1
Reviewer 1 Report
- Line 116, “protein content is determined by the Kjeldahl method.” is seldomly used on the assay of enzyme.
- Line 117, Description on determination of Molecular mass distribution is not clear. Such as, MW marker, assayed machine, conditions for the determination.
- Table 1, what is SPRE (%) ? No information on the determination of Viscosity.
- Table 2, what is PV?
- Line 379 or Table 8, no information on the determination conditions of amino acid using GC.
- Data obtained from current manuscript seems to be not very important in this field.

The introduction should be rewrite because it has nothing to do with the experimental part.
In the methods section:
Preparation of soybean soup should be rewrite. Soup is not scientifically at all.
The Kjedhal method should be described or at least a citation should be added.
The mass distribution section should include the analysis conditions not general infomartions about the GPC separation.
The Figures are blurred and should be improved.
At Figures where the y values are in percents, no digits are needed.
The novelty of the paper is not mentioned.
The results are not correlated with the previously reported results.

This manuscript can be acceptable without major revision.
But, some corrections are required.
- At the bottom of Figure 8, (blue box)Flavourzyme, (yellow box)Alcalase should be included like Figure 10.
- References should be consistent. Journal abbreviation, Author names (i.e, First and Last name)

Round 2

- Line 379 or Table 8, no information on the determination conditions of amino acid using GC.
Determination conditions of amino acid using GC
Equipment: HPLC Agilent 1100 system, Zorbax extend C 18 column (4.6 x
250 mm), UV probe 250 nm.
Mobile phase: phase A: pH 2.8 buffer - Phase B: 50% ACN, 50% IPA:
Flow rate: 1mL / minute - This is a conditions for HPLC not for GC.
- MW maker and HPLC chromatogram were not fonud in the text.

In my opinion the present form of the Figures is not OK.
