Purification, Characterization, and Application of Alkaline Protease Enzyme from a Locally Isolated Bacillus cereus Strain
Round 1
Reviewer 1 Report
In this study, authors investigated the Purification, characterization, and application of alkaline protease enzyme from a locally isolated Bacillus cereus strain. Though some results are interesting, this paper still needs greatly improvements in writing, data presentations and discussion. I suggest after minor revisions, this paper could be considered to accept for publication.
Commments.
1. Authors should provide new version of figure for SDS-page with high quality.
2. Ph Value or pH Value? Pls check the whole text to avoid such errors.
3. Authors should give insightful discussion especially compared with previous reports, not only repeat data obtained in this study.
4. Conclusion section should be rewritten, should more focus on what you obtained in this study and what you will carry out further.

Comments.
- Authors should provide new version of figure for SDS-page with high quality.
AR: Thank you for your comment. A good quality with high resolution figure has been provided in the revised manuscript. Please see revised manuscript.
- Ph Value or pH Value? Pls check the whole text to avoid such errors.
AR: Thank you for your correct it has been corrected in the revised manuscript as pH value. please see revised manuscript.
- Authors should give insightful discussion especially compared with previous reports, not only repeat data obtained in this study.
AR: Thank you for your comment, discussion section has been improved in the revised manuscript. Please see revised manuscript.
- Conclusion section should be rewritten, should more focus on what you obtained in this study and what you will carry out further.
AR: Thank you for your comment, conclusion section has been improved in the revised manuscript. Please see revised manuscript.

Reviewer 2 Report
General evaluation of this manuscript: On the scientific point of view, this work is a contribution, despite not introducing novel concepts, to the field of bacterial alkaline proteases.
These are some specific concerns/suggestions that Authors should address:
- Page 2: Please insert the Enzyme Commission number (EC numbers) the first time that an enzyme used and assayed is referred.
- Page 2 line 54: “Proteases or peptidases are a class of enzymes that classified as proteolytic.” this sentence needs a revision.
- Introduction, lines 47-77: needs a review/rewrite in order to eliminate redundant information, e.g., proteases and global enzyme market.
- Page 2 line 89: “B. subtilis” change to italic.
- Page 3 line 121: “immense” or immersed?
- Page 3 line 122: “Ions of Salt diffused out via membrane.” Please delete (very basic information)
- Page 3 lines 127-131: Please review and simplify column packing description.
- Page 4 line 201: please add a brief methodology description of enzyme activity determination. Also, definition of one unit of enzyme activity (U) is necessary.
- Page 5 line 220: “elusion” or elution?

These are some specific concerns/suggestions that Authors should address:
- Page 2: Please insert the Enzyme Commission number (EC numbers) the first time that an enzyme used and assayed is referred.
AR: Thank you for your comment. Enzyme commission number has been added to the revised manuscript accordingly.
- Page 2 line 54: “Proteases or peptidases are a class of enzymes that classified as proteolytic.” this sentence needs a revision.
AR: Thank you for your comment, this sentence has been changed to “Proteases or peptidases are classified as proteolytic enzymes”.
- Introduction, lines 47-77: needs a review/rewrite in order to eliminate redundant information, e.g., proteases and global enzyme market.
AR: Thank you for your comment. These lines were rewritten and highlighted in the manuscript.
- Page 2 line 89: “B. subtilis” change to italic.
AR: Thank you for your comment. It has been changed in the manuscript accordingly.
- Page 3 line 121: “immense” or immersed?
AR: Thank you for the correction. It has been changed to Immersed. Please see revised manuscript.
- Page 3 line 122: “Ions of Salt diffused out via membrane.” Please delete (very basic information)
AR: Thank you for your comment. It has been deleted in the revised manuscript accordingly. Please see revised manuscript.
- Page 3 lines 127-131: Please review and simplify column packing description.
AR: Thank you for your comment it has been Simplified in the revised manuscript accordingly.
- Page 4 line 201: please add a brief methodology description of enzyme activity determination. Also, definition of one unit of enzyme activity (U) is necessary.
AR: Thank you for your comment. A brief methodology of enzyme activity determination has been added to the revised manuscript. One unit of enzyme activity is equal to 1u mol of amino acid released by the per ml/per minute of enzyme source.
- Page 5 line 220: “elusion” or elution?
AR: Thank you for the correction it has been corrected as elution in the revised manuscript.
